Purification and some properties of valyl ribonucleic acid synthetase from yeast.

چکیده
The mechanism by which an aminoacyl ribonucleic acid synthetase recognizes its transfer ribonucleic acid substrate constitutes one of the central problems in biochemical genetics. Obviously this problem cannot be solved without a thorough understanding of the structure of both enzyme and substrate. Although work on the structure of tRNAr has yielded signal results during the last few years (l-3), very little progress has been made in the elucidation of enzyme structures. On the other hand, several aminoacyl-RNA synthetases have been obtained in highly purified form (4-15), and studies with some of them have revealed interesting information regarding the formation of enzyme-substrate complexes (16-22). In this paper we describe the purification and some properties of one of these enzymes, valyl-RNA synthetase from yeast.

Structure and function of transfer ribonucleic acid. 3. Some properties of a complex between valyl transfer ribonucleic acid synthetase and transfer ribonucleic acid specific for valine.
The formation and some properties of a stable complex between valyl transfer ribonucleic acid synthetase and transfer RNA from yeast have been investigated by three different methods for the isolation of the complex: titration over Sephadex, sucrose gradient centrifugation, and electrophoresis on Pevikon as supporting medium. Transfer RNA specific for valine (tRNAvsl) can be separated into two ...
متن کاملViral Modification of the Valyl Transfer Ribonucleic Acid Synthetase of Escherichia COW
The valyl-tRNA synthetase (EC 6.1.1.9) of Escherichia coli was compared to the modified form of the enzyme which develops during infection by bacteriophage T4. The two forms were found to differ in their stability to heat, stability to urea, ability to charge yeast tRNA, electrophoretic mobility, sedimentation rate in sucrose gradients, and molecular size. Both enzyme activities are easily puri...
متن کاملPurification and properties of a T4 bacteriophage factor that modifies valyl-tRNA synthetase of Escherichia coli.
After T4 bacteriophage infects Escherichia coli, a peptide tau, produced under the control of a phage gene, binds to the host valyl transfer ribonucleic acid synthetase (EC 6.1.1.9) and thereby changes several of its physicochemical properties. The interaction of tau with the host enzyme was investigated in vitro after extensively purifying the factor from T4-infected E. coli using a rapid puri...
متن کاملAffinity chromatography of aminoacyl-transfer ribonucleic acid synthetases. Cognate transfer ribonucleic acid as a ligand.
The use of tRNA affinity columns for the purification of aminoacyl-tRNA synthetases was investigated. A purification method for valyl-tRNA synthetase from Bacillus stearothermophilus is described that uses two affinity columns, one containing the pure cognate tRNA, and the other containing all tRNA species except the cognate tRNA. A method for the rapid preparation of the two columns was develo...
